Tips on manual mode SLR photography?

Recently I've been trying to get out of my comfort zone of using the auto modes on my Nikon D7000 and start using the manual mode instead.It is taking me a long time to get my head around it all. I understand what ISO, aperture and shutter speed do. But it takes me about 5 minutes to get the right balance between them all! I'm just not getting the hang of it at all! I feel like I should KNOW how much of each I need, rather then it being a 5 minute trial and error session.

Are there any helpful tips you could give me?
